Call for Assays: 3D Liver Toxicity Models for the Regulatory Assessment of Chemicals

The DRP aims to:

• Review the state of the art of 3D liver toxicity models relevant for regulatory use;
• Analyse the readiness and maturity of different test systems;
• Identify scientific, technical, and regulatory challenges and opportunities;
• Support future development and potential regulatory acceptance of in vitro liver toxicity methods.

In this context, the purpose of the Call for Assays is to:

• Obtain an overview of existing and emerging 3D liver toxicity models;
• Understand their intended purpose, what they measure, their development status, and potential relevance for regulatory toxicology;
• Support the DRP section addressing the readiness of test methods for regulatory applications, with a particular focus on non‑pharmaceutical chemicals (e.g. industrial chemicals, pesticides, biocides, cosmetics).

The scope of the call is broad and covers 3D liver models, including organoids, spheroids, and organ‑on‑chip systems, using test systems based on iPSC‑derived cells, primary cells, or immortalised cell lines.

While the DRP places particular emphasis on applications for non‑pharmaceutical chemicals, models originally developed for medical or pharmaceutical purposes are also considered relevant in this Call for Assays, where they can be adapted for use in other areas of regulatory toxicology.

Methods at different stages of development are welcome.
