In 2022, Test Guideline 456: H295R Steroidogenesis Assay was revised and a new data interpretation procedure (DIP) adopted. This new DIP introduced a new (additional) criteria to conclude to a positive result (induction or inhibition), i.e. result > 1.5-fold induction or inhibition threshold from the solvent control. It recently appeared that this new way of assessing the results leads to changes in the expected outcome of some proficiency chemicals. Therefore, the TG needs to be adapted to ensure that the results specified for the recommended proficiency chemicals all align with the expected outcomes using the new data interpretation procedure.

The draft corrected TG 456 is currently under a commenting round by the Working Party of the National Coordinators of the Test Guidelines Programmes and open for comments from the public at the same time. Please note that only the proposed changes are open for comments. Interested parties are invited to contact their National Coordinators at least a week before 23 January 2025.
